چکیده

In the course of a study of the blood coagulation reaction involving the use of partially purified reagents it was thought of some interest to determine effects of fibrinolytic enzyme on these isolated materials. It is well known that serum lysin (fibrinolysin, plasmin) will lyse or destroy both fibrinogen and fibrin. It may be thrombinolytic (1). Seegers and Loomis (2) have stated that it destroys prothrombin. In the present study we have compared the effects of lysin on prothrombin, Ac-globulin and tissue thromboplastin. In choosing a lysin for these studies it was necessary to find a preparation active fibrinolytically but free from thrombin and antithrombin. A preparation of Loomis' (3) fibrinolysin was satisfactory in these respects. Human Fraction III-3 (4) and our dog serum lysin (5) both contained appreciable quantities of thrombin.

Studies on a Proteolytic Enzyme in Human Plasma

A fraction of globulin was prepared from human plasma which was deficient in prothrombin, thrombin, fibrinogen, plasma thromboplastin, and accelerator globulin. The preparation of globulin contained considerable potential proteolytic activity which could be activated by streptococcal fibrinolysin. This fraction of globulin accelerated the clotting of normal platelet-deficient plasma. However, t...

The Prothrombin Conversion Accelerator of Serum

The conversion of prothrombin to thrombin during normal coagulation proceeds with ever increasing velocity until at the moment of clotting, or shortly thereafter, thrombin evolution is explosive. This phenomenon, referred to as the "autocatalytic" nature of thrombin formation, remains unexplained. It is evident that abnormalities in this reaction may underlie the pathogenesis of certain hemorrh...

Studies on prothrombin; purification, inactivation with thrombin, and activation with thromboplastin and calcium.

Prothrombin, thrombin, fibrinogen, thromboplastin, Ac-globulin,’ librinolysin, antifibrinolysin, antithrombin, antithromboplastin, antihemophilia globulin, and heparin are all factors which are known to be a part of the blood clotting mechanism. They function either to provide emergency hemostasis or to prevent intravascular clotting. In the laboratory their activities are recognized and measur...

A Factor in Serum Which Accelerates the Conversion of Prothrombin to Thrombin: I. Its Determination and Some Physiologic

INTRODUCTION I N 1945, Nolf1 described a serum constituent, distinct from thrombin, which was capable of furthering coagulation. More recently, Ware et al. reported a substance, designated serum Ac globulin, which activates the conversion of prothrombin to thrombin by thromboplastin. Independently, Owren5 discovered a new clotting factor, ‘ ‘Factor VI,’ ‘ which arises during coagulation, speeds...

ANALYTICAL REVIEW Conversion Factors and Accelerators in the Formation of Thrombin

E VEN to the reader intimately acquainted w’ith the field, several of the recent developments in the study of the mechanism by which blood clots have often proved extremely puzzling. The number of communications which have appeared on the subject , especially during the past ten years, is indeed amazing, and their quantity alone may ‘el1 preclude the possibility of fair critical appraisal. More...
